Not really. The AP Bio textbook does go pretty in depth. I took the test at Nats with one day's notice (one of the captains couldnt come, long story) so I studied the AP Bio textbook, cause I was in the class.
And most of the stuff was in the book. Sure I didnt remember it all because I studied it for like 2 hours, but for people with more warning, studying the chapters on cells is plenty.

Here's a more specific question:
How is the test formatted? All multiple-choice, or are there short answer sometimes? I would imagine it would vary by test writer, but in general.

That's it. It's going to depend on who writes the test for the competition. There's really no standard among SO tests.

Bioethics is a new topic on the rules. How much / indepth do you think it'll be? Most bio books only briefly mention the topic in the chapter for bioengineering.

I'd say it would be in pretty much no depth until May 20-22.

On one of my last invitational tests, it listed a series of DNA sequences and asked which was the most stable. How do you tell if a sequence is stable? There were no issues in matching base pairs or stuff like that.
